Screening Differential Expression Profiles of Urinary microRNAs in a Gentamycin-Induced Acute Kidney Injury Canine Model

نویسندگان

چکیده

microRNAs (miRNAs) are promising biomarkers for different pathological models because of their stable and detectable characters in biofluids. Here, we collected urine samples from 5 beagle dogs on the 3th, 6th, 12th day an acute kidney injury (AKI) caused by gentamycin. miRNA levels were measured with high-throughput sequencing results then differentially investigated. Gene Ontology (GO) KEGG pathway analysis performed to analyze potential target genes corresponding expressed miRNAs (DE-miRNAs). Relationships between hub DE-miRNAs analyzed STRING Cytoscape. We identified 234 3, 6, 12 days after gentamycin treatment (p < 0.05). Top 10 up- down-regulated candidate predicted overlapping TargetScan miRanda results). GO analyses demonstrated that mainly involved injury-related pathways, such as insulin signaling pathway, oxytocin hedgehog pathway. The network miRNA-hub suggests miR-452, miR-106a, 106b participate regulating largest number genes. evaluated signature via a canine model built gentamycin-caused injury. Our represent valuable resource evaluating renal toxicity.

MicroRNAs in Pathogenesis of Acute Kidney Injury.

MicroRNAs (miRNAs) are small non-coding RNAs that regulate gene expression mainly by repressing their target gene translation. A large spectrum of human diseases is associated with significant changes in miRNAs. Many miRNAs are induced in diseases, whereas some others are downregulated. The significance of miRNAs has been demonstrated in renal development and physiology, and in major kidney dis...

MicroRNAs in the kidney: novel biomarkers of acute kidney injury.

microRNAs are small, endogenous RNA molecules which are critical for a new step in the regulation of the gene expression. They have become the most critical biological mediators characterized in the last ten years. microRNAs participate in almost every cellular process, therefore their deregulation is associated with the development of a wide range of pathologies, including kidney diseases. Inc...

Wasp stings (Vespa affinis) induced acute kidney injury following rhabdomyolysis in a 25-year-old woman.

Wasp sting is a relatively common arthropod assault. This usually results in pain and mild allergic reactions, but sometimes may cause severe systemic reaction and multiorgan dysfunction including rhabdomyolysis, hemolysis, coagulopathy, hepatic, renal and cardiac complications. Along with several other pathomechanisms, rhabdomyolysis is a distinguished cause of acute kidney injury (AKI) in pa...

Differences in gene expression profiles and signaling pathways in rhabdomyolysis-induced acute kidney injury.

PURPOSE Rhabdomyolysis is a threatening syndrome because it causes the breakdown of skeletal muscle. Muscle destruction leads to the release of myoglobin, intracellular proteins, and electrolytes into the circulation. The aim of this study was to investigate the differences in gene expression profiles and signaling pathways upon rhabdomyolysis-induced acute kidney injury (AKI). METHODS In thi...

Acute Ozone-Induced Differential Gene Expression Profiles in Rat Lung

Ozone is an oxidant gas that can directly induce lung injury. Knowledge of the initial molecular events of the acute O3 response would be useful in developing biomarkers of exposure or response. Toward this goal, we exposed rats to toxic concentrations of O3 (2 and 5 ppm) for 2 hr and the molecular changes were assessed in lung tissue 2 hr postexposure using a rat cDNA expression array containi...
